Total flavonoids from Saussurea involucrata attenuate inflammation in lipopolysaccharide-stimulated RAW264.7 macrophages via modulating p65, c-Jun, and IRF3 signaling pathways

Objective: To investigate the anti-inflammatory effects of total flavonoids from Saussurea involucrata on lipopolysaccharides (LPS)-stimulated murine RAW264.7 macrophages and explore its underlying mechanism action. Methods: Total were extracted using chromatographic column method. Cell viability was determined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide assay. The production nitric oxide detected Griess assay release cytokines (IL-10 TNF-α) chemokines (MCP-1, MIP-1a, CCL5/RANTES) ELISA to evaluate activity involucrata. Moreover, nuclear translocation p65, c-Jun, IRF3 immunofluorescence microscopy Western blotting analysis performed determine expression related proteins. Results: 751.5 mg/g content rutin 506.5 mg/g. inflammatory mediators including oxide, cytokines, effectively inhibited Meanwhile, also suppressed in LPS-stimulated cells. LPS-induced iNOS COX-2 remarkably reduced treatment with decreased levels p-IKKa/β, p-TBK1, p-p38, p-ERK, p-JNK, p-p65, p-c-Jun, p-IRF3 LPS-exposed macrophages. Conclusions: potentially inhibit secretion pro-inflammatory mediators, which may be inhibition signaling pathways
